Macdonald (Mac) Christie is Professor of Pharmacology and Associate Dean (Research) at Sydney Medical School and a NHMRC Senior Principal Research Fellow at the University of Sydney. He is a neuropharmacologist investigating the cellular and molecular mechanisms of opioid receptor signaling in pain pathways, the biological basis of adaptations that produce chronic pain and drug dependence, and is involved in preclinical development of novel pain therapeutics derived from conotoxins. Prof Christie is internationally renowned for his studies on ion channel function and neural network plasticity in nervous system disorders such as chronic pain and drug dependence. After completing his PhD at the University of Sydney, he spent a number of years as a postdoctoral research scientist – including Fogarty International Fellow at the Massachusetts Institute of Technology and subsequently, Senior Postdoctoral Fellow at the Vollum Institute in Oregon, USA. He returned to the University of Sydney as a tenured academic at the Department of Pharmacology and was appointed Professor (Personal Chair) in 1999. Prior to his present appointment, Prof Christie held various senior leadership roles at the University and its affiliated institutes including Head of Department of Pharmacology at the University of Sydney, Director (Basic Research) and Professor at the Pain Management Research Institute, Royal North Shore Hospital, and Director of Pain and Addiction Research at the Brain and Mind Research Institute, University of Sydney. He has published over 200 original research articles in the neuroscience field, with several publications in highest-ranked journals including Nature, Science, PNAS, Nature Neuroscience, Neuron, Journal of Neuroscience, and Pain and he regularly reviews manuscripts for many of these journals. He is the author of invited reviews to major journals such as Pharmacological Reviews, the British Journal of Pharmacology and Lancet Oncology. His publications have received >11,000 citations (GS H-Index = 58). Prof Christie has received continuous grant funding as lead investigator from the NHMRC since 1991 and from other bodies including ARC and NIH with a total of > AUD 9 million in competitive funding received since 2008. Currently, he is Chief Investigator on a NHMRC Program Grant. He actively supports and promotes research excellence – he is a member of the Board of Directors of the Woolcock Institute of Medical Research and of the Sydney Cancer Institute. He is a Council member of the Sydney Medical School Foundation, member of the University Senior Executive Group Research Committee, Chair of the Sydney Medical School Research Committee, and member of the NSW Tissue Resource Centre Management Committee. In addition, he has been a long-standing contributor to a number of international research agencies including since 2001 as member of the Human Frontier Science Program scientific review panel and scientific referee for the Wellcome Trust (UK), and with the National Science Foundation (USA) since 2005.

Prof Christie serves on editorial boards of a number of prestigious international journals including the British Journal of Pharmacology and he is a past member of the Executive of the International Narcotics Research Conference, the peak international opioid research organization. Additionally, he assumes a senior mentorship role for postdoctoral fellows at the university. Throughout his career, he has been involved in the development of excellence in postgraduate training, having served on the Faculty of Medicine’s Board of Postgraduate Studies (including as Deputy Chair), as Chair of Faculty and College Scholarships committees and by promoting matters relating to postgraduate education on the Faculty Research Committee. Prof Christie also consults/makes submissions to government on matters of policy including in the past, the analysis and reporting to NSW Health on the pharmacology of psychostimulants and on high-dose methadone therapy, advice to the NSW Police Integrity Commission on drug and alcohol testing of NSW Police, and as a member of the NSW Premier’s Cabinet Working Party on the medicinal uses of cannabis (2001).

Research interests

Prof Christie is an internationally renowned electrophysiologist and expert in ion channel and synaptic physiology and pharmacology and the leading basic opioid pharmacologist in the nation. His most important achievements relate to studies on the molecular nature and integrated function and plasticity of signalling elements in neurons, which contribute to chronic pain and opioid tolerance/dependence, and his work on the development of potential therapeutics for pain. His research has heralded numerous ‘firsts’ – early in his career, Prof Christie contributed to the first cloning of the D2 dopamine receptor (published in Nature) and achieved the first expression of a mammalian voltage-gated potassium channel (published in Science), and first functional demonstration of its heteropolymerisation (published in Neuron). His later research uncovered the mechanisms of plasticity of G-protein receptor coupling in neurons that are transforming thinking regarding opioid tolerance and ion channel plasticity in chronic pain states. His group discovered that µ and δ opioid receptors in nerve cells share common G-Protein receptor coupling mechanisms and provided the first evidence of direct coupling of µ-receptors to GIRK channels and of coupling of the opioid-related receptor ORL1, to GIRK channels in brain as well as in cannabinoid signaling. He discovered a novel signalling mechanism for G-Protein Coupled Receptors in synapses in brain (published in Nature). This provided a basis for seminal findings on plasticity of signaling in opioid withdrawal published in Neuron (2005) and recently in Nature Neuroscience (2011).

Currently, Prof Christie continues to undertake research on opioid receptor regulation in pain and analgesia, in particular the processes driving the development of opioid tolerance. His team is studying the molecular mechanisms responsible for tolerance at the µ-opioid receptor, investigating action-related learning and plasticity in the cortico-striatal network, and studying novel GPCR mechanisms to treat neural hyper-excitation in chronic pain. These studies on the mechanisms involved in intracellular signal bias aim to develop novel therapeutics for pain and addiction that could potentially preferentially divert signaling toward pathways associated with increased analgesic efficacy, lower rates of tolerance and/or fewer adverse effects. Additionally, as a Chief Investigator on a NHMRC Program Grant, Prof Christie is leading collaborative research (with researchers at the University of Queensland and RMIT in Melbourne) on the development and discovery of novel analgesics based on conotoxins. The research teams have discovered a number of conotoxin peptides that selectively block pain pathways without producing tolerance, which are currently being characterised further for improved pain relief in humans.
