Biographical details

Patrick Bertolino completedhis PhD in France (Lyon University)beforeundertaking post-doctoral studies at the Walter and Eliza Hall Institute in Melbourne, at the ENS-Lyon in France and attheCentenary Institute in Sydney where he established the Liver Immunology group in 2001. He has beena Faculty member of the Centenary Institute since 2011. He hasmade some seminal discoveries in this fieldand isinternationallyacknowledged as one ofthetop leading experts in Liver Immunology worldwide. These discoveriesincludethefirst reportthat naïve CD8 T cells can be activated directly in the liver, a fundamental finding that overturned the conventional belief that primary T-cell activation is unique to lymphoid tissues.This research is important in several clinical settings including transplantation, malaria and chronic hepatitis C virus (HCV) infection.

Research interests

The liver Immunology Groupusesunique transgenic mouse models, advanced imaging technology and flow cytometryto dissect mechanisms regulating the balance between tolerance and immunity in the liver andhow T cells interact and are instructed by hepatic cells in both the healthy and diseased or transplanted liver.This research is important in transplantation, malaria and chronic hepatitis C virus (HCV) infection. The groupfocuses ontwocurrentoutstanding questions in Liver Immunology:

A)What are the mechanisms by which the liver induces tolerance to avoid acute injury? The liver is known to induce antigen-specific tolerance but mechanisms remain unknown. the group has shown that the hepatic microvasculature allows the liver to act as a site of primary T cell activation, a property thought to be restricted to 2º lymphoid tissues. In contrast to activation in lymphoid tissues, intrahepatic activation leads to tolerance. The group has recently demonstrated that most liver-activated T cells invade hepatocytes and are rapidly destroyed in lysosomes.

B) What are the parameters allowing an acute liver injury to become chronic? The liver is a site of many chronic infections but the parameters allowing progression to chronic hepatitis are unclear. The group has developed unique models of acute immune mediated hepatitis to dissect mechanisms leading to chronic hepatitis.

Current projects

The research of the Liver Immunology group focuses on 4 main projects :

1-Understand the parametersthat determine the balance of immunity andtolerance in the liver.

2- Determine the molecular pathways and the physiological relevance of T cell invasion and degradation in hepatocytes.

3-Dissect the early events of T cellactivation leading to tolerance inliver transplantation.

4-Characterise the parameters leading to chronic mediated hepatitis.
