My research interests and expertise centres on the inorganic chemistry and chemical biology of bacterial metabolites known as siderophores that contain hydroxamic acid functional groups with high affinity towards complexing Fe(III) and other metal ions relevant to biomedicine, and metal recovery. My interdisciplinary research blends chemistry (inorganic, organic, and medicinal), molecular biology and biotechnology. I am internationally renowned for innovations in the design and production of metal binding compounds (termed ‘chelators') for imaging/therapy and/or managing metal dyshomeostasis and/or environmental remediation/recovery. My team has: (i) improved hydroxamic acid syntheses; (ii) produced using Zr(IV)-based metal-templated synthesis two new chelator chemotypes with potential for Zr(IV)-89 immunological positron emission tomography (PET) imaging; (iii) produced new water soluble Zr(IV)-89 chelators using a combined microbiological-semi-synthetic chemistry approach; (iv) invented a first-in-class dual-chelator to enable the development of immunological theranostic agents; and (v) pioneered a chemo-enzymatic approach to deliver biocombinatorial pools of multimeric metal chelators to reveal new Ga(III) and Zr(IV) coordination chemistry. In parallel, my team conducts research on bacterial metabolomes for siderophore discovery and to inform biosynthetic pathways, and the design and production of iron chelators with the aim to expand options to manage transfusion-dependent secondary iron overload disease. Collaborators include: QIMR Berghofer Medical Research Institute, The Department of PET and Nuclear Medicine (RPAH), The University of Melbourne, the Florey Institute of Neuroscience and Mental Health, the Children's Cancer Institute, Australian Nuclear Science and Technology Organisation (ANSTO), Centre for Advanced Imaging, University of Queensland, Telix Pharmaceuticals, University of New South Wales, CSIRO.
I have a major teaching presence into PCOL3012/3912 Drug Design & Development and also teach into MCHM3001/3901 From Molecules to Therapeutics. I am the academic lead for the major in Medicinal Chemistry in the Bachelor of Science and Bachelor of Advanced Studies.
