Thesis title: Mechanisms of gene regulation

Thesis abstract:

Nucleosome remodeling and deacetylase (NuRD) complex is an important epigenetic regulator of chromatin structure and gene expression. NuRD complex is an assemblage of proteins that combine key epigenetic regulators necessary for (1) histone deacetylation and demethylation, (2) binding to methylated DNA, (3) mobilization of nucleosomes, and (4) recruitment of additional regulatory proteins. NuRD complex is essential for embryonic development and pluripotent stem cell differentiation. It has also been implicated in the regulation of transcriptional events integral to oncogenesis and cancer progression.Emerging molecular details regarding recruitment of NuRD to specific loci during development and modulation of these events in cancer are used to illustrate how inappropriate localization of the complex could contribute to tumor biology. The goal of this project is to determine the mechanistic basis for the activity and selectivity of the nucleosome remodeling and deacetylase (NuRD) complex, a co-regulator that is strongly linked to cancer. The size, complexity and low abundance of these complexes means that relatively little is known about the molecular mechanisms underlying their activity.
