Biographical details

Ann graduated with a PhD in biochemistry from the University of Sydney in 2005 under the supervision of Prof Joel Mackay. Following her PhD, she worked in Prof Mingjie Zhang's laboratory at the Hong Kong University of Science and Technology and in Prof Jacqui Matthews' laboratory at the University of Sydney as a postdoctoral research associate. In 2006, Ann was awarded the Australian Postdoctoral Fellowship from the Australian Research Council which has allowed her to embark on an independent research career studying the structures of a class of unique self-assembling proteins from fungi which are involved in coating spores and mediating fungal infections. She has been awarded three younger investigator awards for her work.

In 2009, coinciding with the establishment of the 800 MHz NMR Facility which houses the then highest field NMR spectrometer in NSW, Ann took up the position of NMR Facility Manager at the former School of Molecular Bioscience at the University. This has allowed Ann to expand her NMR skill base to include small molecule, metabolic and solid-state NMR methods.

In 2012, Ann was appointed a Research Fellow and runs a group that focuses on protein self-assembly, protein:protein interactions, and more recently fragment-based drug design at the School of Life and Environmental Sciences (SOLES). She has also developed an interest in NMR metabolomics through a number of collaborative projects.

Ann is currently a Senior Research Fellow at SOLES.

Research interests

Ann's lab is focused on solving interesting biological problems using a cross-disciplinary approach.

The research areas we work on include: protein engineering using functional amyloids, drug design to fight antibiotics resistance and gaining a molecular understanding of diseases.

One of her core research interests is a class of fungal proteins called hydrophobins. These proteins can self-polymerise to form a strong coating on structures that can reverse surface polarity. In fungi, hydrophobin coatings confer water resistance to spores and aerial hyphae and are also responsible for mediating interactions with host surfaces to initiate infections. In some cases, the coatings have amyloid characteristics and additionally raise the question of how biological function can be compatible with the amyloid scaffold. Additionally, Ann and her team are interested in expanding the functionality of hydrophobin coatings through “smart” protein engineering to promote the adhesion and growth of osteoblasts in PEEK bone implants.

This work is a collaboration with A/Prof Sunde from the School of Medical Sciences and A/Prof Suchowerska from the Chris O'brien Lifehouse.

The other key focus of Ann's lab is to develop new classes of antibiotics by targeting specific molecular interactions in bacteria. The two systems her lab works on are 1) the bacterial Signal Recognition Particle (SRP) and its Receptor (SR) from E. coli and 2) the Heme uptake system protein A (HusA) from P. gingivalis. The SRP/SR work is in collaboration with Dr Sandro Ataide at the School of Life and Environmental Sciences while the HusA work is in collaboration with Dr Gao at the Westmead Centre for Oral Health and Dr Gell from University of Tasmania.

More recently, Ann has embarked on a number of metabolomics projects including a collaboration with A/Prof Suchowerska and Prof McKenzie from the Chris O'brien Lifehouse and School of Physics to investigate cellular signaling in novel cancer radiotherapies.

Teaching and supervision

Ann is a member of the SOLES Honours committee and coordinates the Structrual Biology stream of the Honours program.

Undergraduate Teaching:

Protein Bioinformatics practical class for third year BCHM3x81. This practical class consists of two tutorials and two half days of laboratory sessions.

Advanced module of the third year BCHM3982 course on Biological Problem Solving using NMR spectroscopy. The NMR course consists of eight lectures, one tutorial and five days of laboratory classes.
